Description and Objective

The scope of the proposed ThyroTransVal project is the validation of an in vitro method to identify potential endocrine disruptors of thyroid hormone transmembrane transport. This method utilizes MDCK-1 cells stably transfected with the human thyroid hormone transporter MCT8 (Monocarboxylate Transporter 8). The method enables the investigation of active T3 transport into target cells and its potential inhibition by substances. Thyroid hormone uptake into target cells is measured spectrophotometrically using the Sandell-Kolthoff method. The project's goal is to transfer this method from the developer laboratory (BfRshort forGerman Federal Institute for Risk Assessment) to other laboratories and verify its reproducibility through a blinded ring trial. Successful transfer and validation will be documented in a validation report and reviewed by external experts with the goal to be adopted as an OECD test guideline. The project is structured into three phases over a period of three years: Phase 1: Final characterization of the test system for deposition in a cell bank as well as potential adaptation to an FCS-free medium & transfer of the method to four partner laboratories. Phase 2: A ring trial with 20–30 blinded test substances. Phase 3: Submission of the method as an OECD test guideline based on the validation report.
