Description and Objective

TKplate 2.0 is an extension of TK-Plate, an EFSAshort forEuropean Food Safety Authority open-source platform that integrates physiologically-based kinetic (PBK), physiologically-based kinetic dynamic (PBKD), toxicokinetic-toxicodynamic (TK-TD) and dynamic energy budget (DEB) models for humans and food-producing animals. It provides a user-friendly graphical interface through which these models can be applied for the risk assessment of pesticides, food additives, feed additives and contaminants. There is a need to further develop these models, not just for single chemicals but also for multiple chemicals and biological stressors in farm and wild mammalian species as well as in birds. For this, more knowledge is needed of the toxicokinetic and toxicodynamic processes that link chemical exposureExposureTo glossary to adverse health outcomes in different farm and wild mammalian and bird species. Innovative, new approaches to predict the kinetics and dynamics of chemicals and support inter-species extrapolations are also needed. These may include allometric scaling, QSAR, machine learning and deep learning models to predict parameters such as partitioning, metabolism, kinetic parameters, bioaccumulation, toxicity, substrate-enzyme and/or substrate-transporter interactions. The effect of biological stressors and welfare conditions on the different TK and TD processes in these models must be integrated. The objective is to further develop biologically-based models for risk assessment and address more complex questions on single chemicals, multiple chemicals, biological stressors and welfare conditions in species of relevance to food and feed safety.
