Description and Objective

The majority of chemical carcinogens, or metabolites thereof, induce DNA damage and can be detected by in vitro or in vivo genotoxicity assays. However, some substances may induce cancer without being detectable in currently applied genotoxicity assays, namely non-genotoxic carcinogens (NGTxCs).These might act for instance as tumor promoters, modulators of nuclear receptors as well as inducers of tissue-specific toxicity or immune and inflammatory responses. Here, the assessment of a potential hazard from chemical exposureExposureTo glossary that might lead to cancer relies on long-term rodent-based bioassays that are not only time-consuming, costly and require very high numbers of animals, but, at the same time, are questioned due to limited human relevance. Therefore, there is an urgent need for a transition to in vitro and more human-relevant approaches that exploit our current understanding of cancer and employ the latest non-animal methods available in a modern safety assessment toolbox. Here we describe a project that aims at improving and implementing various novel test systems that address different aspects of carcinogenic mechanisms. With these methods the data will be generated that is needed for the development of an efficient and reliable testing and assessment strategy or defined approaches to ensure a sensitive and specific detection of non-genotoxic carcinogenic compounds.
