werden gewebespezifische Gene, Proteine, Ionenkanäle und
Rezeptoren in einem differenziellen Muster exprimiert, das
dem der Maus-Embryogenese
in vivo
sehr ähnelt. Aufgrund
dieser Eigenschaften werden Stammzellen der Maus bereits
für pharmakologische Untersuchungen und sicherheitstoxi-
kologische Prüfsysteme verwendet. Ein Beispiel ist der am
BfR entwickelte Embryonale Stammzelltest (EST), bei dem
embryonale Stammzellen der Maus zu schlagenden Kardio-
myozyten differenzieren. Der Test ist durch das europäische
Validierungszentrum ECVAM wissenschaftlich validiert und
kann das embryotoxische Potenzial einer Substanz mit einer
Genauigkeit von 78 % vorhersagen.
Im Jahr 2012 hat das BfR die Entwicklung neuer stammzell-
basierter Testverfahren vorangetrieben. Die Arbeiten hier-
zu fanden mit Partnern aus Industrie und Universitäten im
Rahmen mehrerer Verbundprojekte statt, die das Bundes-
ministerium für Bildung und Forschung förderte. Die neuen
Testverfahren eignen sich zur Prüfung auf Embryotoxizität,
Entwicklungsneurotoxizität und Entwicklungsosteotoxizität.
Dabei wurden zum einen neue gewebespezifische Differen-
zierungsprotokolle für Nerven- und Knochenzellen, die sich
zur Testung von Chemikalien eignen, entwickelt und opti-
miert. Zum anderen wurden neue prädiktive Biomarker zur
toxikologischen Endpunktbestimmung etabliert. Da zur bes-
seren Übertragbarkeit auf den Menschen verstärkt humane
Systeme gefragt sind, konzentrierten sich die experimentel-
len Untersuchungen auch auf die Anwendung von humanen
mesenchymalen Progenitorzellen als Modellsystem für die
embryonale Knochenentwicklung.

is regarded as a model for the development of various
embryonic tissues. During the differentiation, tissue-specific genes,
proteins, ion channels and receptors are expressed in a differen-
tial pattern which is very similar to that of mouse embryogenesis
in vivo
. Due to these properties, mouse stem cells are already being
used for pharmacological studies and in toxicological safety testing
systems. An example is the Embryonic Stem Cell Test (EST) devel-
oped at the BfR which uses the differentiation of mouse embryonic
stem cells into beating cardiomyocytes as an endpoint for develop-
ment. The test was scientifically validated by the European valida-
tion centre ECVAM and can predict the embryotoxic potential of a
substance with an accuracy of 78 %.
In 2012 the BfR promoted the development of new stem cell-based
test methods. The studies were conducted with partners from indus-
try and universities within the scope of several collaborative projects
funded by the Federal German Ministry of Education and Research.
The new test methods are suitable for testing for embryotoxicity,
developmental neurotoxicity and developmental osteotoxicity. New
tissue-specific differentiation protocols for nerve and bone cells suit-
able for the testing of chemicals were developed and optimised.
Furthermore, new predictive biomarkers for toxicological endpoint
determination were established. As more and more human systems
are being in great demand to improve transferability to humans,
experimental testing was also focused on the use of human mes-
enchymal progenitor cells as a model system for embryonic bone
development.
