Description and Objective

Goal of the EDCMET project is to develop new and improved methods for the identification and characterization of metabolic endocrine disrupters. This comprises in silico, in vitro and in vivo methods. BfRshort forGerman Federal Institute for Risk Assessment contributes to the project specifically by the development of test systems to characterize the steatotic potential of chemicals and the activity of xenobiotic-activated nuclear receptors, which play a decisive role not only in the development of steatosis, but constitute also important molecular switches in various other toxicological mechanisms of action. All in all, the project is aimed at contributing new methods to identify metabolic endocrine-active chemicals in a regulatory context, in order to thus improve consumer health protection.

Result

The project was finalized successfully. Test compounds with known or suspected metabolic endocrine activities were selected and characterized using different test systems. The contribution of BfRshort forGerman Federal Institute for Risk Assessment was human liver cell-based test systems for the determination of nuclear receptor activation, transcriptional changes and steatosis induction. The methods were developed further and optimized throughout the project. The test method for the determination of triglyceride accumulation in HepaRG cells was successfully put forward until the prevalidation phase. Prevalidation, with the goal of future validation and regulatory adoption of the test system, is continued within the framework of the PEPPER initiative. A comprehensive protocol of the methods for AOP-based steatosis testing was published.
